The race, which took place in scorching conditions, was highlighted by an eight-man break until Du Plooy and Venter broke clear towards the end.
On Saturday, Du Plooy also won the U/23 time-trial title ahead of Jarrod Hattingh (LEADout-Spier) and Bronwin Adams (Alfa Bodyworks).
The elite race against the clock was won by Tennent with Theuns van der Bank (Giant) and Nabigh Davids second and third.
