The OfficeGuru rider sprinted ahead of Clint Hendricks of Team Enza, while ProTouch rider Jayde Julius completed the podium.
Fourth place went to Louis Visser, with Jaco Venter completing the top five.
“It was a very long sprint and I could sense Clint coming up beside me, but I held on,” said Barrett.
“I still can’t believe it. Winning the Cycle Tour is what every rider dreams of.”
